TMET-26. PRIMARY OLIGODENDROGLIOMA CELL CULTURE VIABILITY: AN IN VITRO STUDY WITH METABOLIC MODULATORS

Abstract Oligodendrogliomas are tumors that develop from oligodendrocytes, the myelinating cells of central nervous system. Oligodendrocytes highly metabolically active synthesize and transfer metabolites to neighboring cells. Given its intimate metabolic relation with neurons, we aim investigate oligodendrocyte metabolism as an antitumoral target. A recurred oligodendroglioma WHO grade III was surgically removed a 50-year patient after 10 years progression-free disease. The tumor sample mechanically digested cultured at 37oC, 5% CO2, in DMEM: F12, % FBS antibiotics. Primary were trypsinized seeded 96-well plate. After 24 hs, treated modulators: metformin (MET, mitochondrial complex II inhibitor, 5 mM), 2 deoxyglucose (2DG, hexokinase 1 6-aminonicotinamide (6AN, pentose phosphate pathway 50 µM) and/or 1400W S-methylisothiourea (both iNOS inhibitors, µM, µM respectively).
